Abstract:

Objective

To investigate the effect of regulatory protein tyrosine and tryptophan hydroxylase activator ζ (YWHAZ) on the proliferation of renal tubular epithelial cells under normal oxygen conditions and hypoxic-reoxygenation conditions.

Methods

Using human proximal renal tubular epithelial cell line (HK-2) through RNAi technology and plasmid transfection to establish a model of low expression and overexpression of YWHAZ, observe the general situation of cells after intervening YWHAZ expression, and detect cell proliferation activity; Then give cells hypoxia-reoxygenation treatment to detect changes in cell proliferation.

Results

Successfully established YWHAZ low and overexpression cell models. After low expression of YWHAZ, the proliferative capacity of cells decreased compared with the control group, and the proliferative capacity of cells decreased more significantly after hypoxia-reoxygenation treatment; Overexpression of YWHAZ alleviates the inhibition of cell proliferation caused by hypoxia-reoxygenation.

Conclusion

YWHAZ may be an important factor for maintaining the proliferative activity of proximal tubular epithelial cells.

Key words: YWHAZ, Hypoxia-reoxygenation, Renal tubular epithelial cells
