Abstract:
Objective To explore the peripheral blood mononuclear cells (PBMC) expression of PPARγ and NF-κB mRNA and its correlation with cardiovascular remodeling in patients with hypertensive renal injury.
Methods From May 2016 to May 2019 in our hospital, 65 patients with hypertension were selected as the simple hypertension group, and 50 patients with hypertensive renal injury were selected as the hypertensive renal injury group, while 50 healthy persons were selected as the control group. The expression of PPARγ mRNA and NF-κB mRNA in PBMC was measured. Cardiac and vascular ultrasound was used to detect the remodeling indicators of carotid artery and heart: carotid artery mean intima-media thickness (IMT), carotid plaque score, and left ventricular mass index (LVMI). Pearson analysis was used to analyze the correlation between the expression of PPARγ and NF-κB mRNA and cardiovascular remodeling indexes. Multiple linear regression method was used to analyze the influencing factors of IMT in the patients with hypertensive renal injury.
Results Compared with those in the control group, the IMT, plaque score, LVMI, and NF-κB mRNA expression were significantly higher in the simple hypertension group and hypertensive renal injury group, while the PPARγ mRNA expression was significantly lower (P<0.05). The IMT, plaque score, LVMI, and NF-κB mRNA expression in the hypertensive renal injury group were significantly higher than those in the simple hypertension group, while the PPARγ mRNA expression was significantly lower than that in the simple hypertension group (P<0.05). In the hypertensive renal injury group, the expression of PPARγ mRNA was negatively correlated with IMT, plaque score, and LVMI (P<0.05), but the expression of NF-κB mRNA was positively correlated with IMT, plaque score, and LVMI (P<0.05). The decrease of PPARγ mRNA and the increase of NF-κB mRNA were the influencing factors of IMT in patients with hypertensive renal injury (P<0.05).
Conclusion The expression of PPARγ mRNA in PBMC of patients with with hypertensive renal injury was significantly decreased, while the expression of NF-κB mRNA was significantly increased. Both of them were correlated with the cardiovascular remodeling.
